Daily increment validation in fish otolith is fundamental to studies on fish otolith microstructure, age determination and life history traits, and thus is critical for species conservation and fishery management. However, it has never been done for Schizothoracine fish, which is the dominant component of fish fauna in the Tibetan Plateau. This study validated the daily increment formation of Gymnocypris selincuoensis, as a representative of Schizothoracine fish, by monitoring the growth of hatchery‐reared larvae group and wild‐caught post‐yolk‐sac larvae group under controlled experiments. The results from monitoring the hatchery‐reared larvae group showed that sagittae and lapilli were found in yolk‐sac larvae, and formed 5–7 days before hatching, but asterisci were not found until 11 days post‐hatching. The first increment in sagittae and lapilli was formed in the first day after hatching. The daily periodicity of increment formation was examined and confirmed in sagittae and lapilli of both larvae groups. However, sagittae were better for age determination than lapilli for larvae at earlier days. For larval G. selincuoensis older than 50 days, lapilli were the only otolith pair suitable for larvae daily age determination. This study validated the daily increment formation in Schizothoracine fish for the first time has primary implications to other fishes from this subfamily.  相似文献

青海湖裸鲤肠道乳酸菌多样性与抑菌活性   总被引:1,自引:0,他引:1  
【目的】通过生理生化特性,结合16S r RNA基因序列分析研究青海湖裸鲤肠道乳酸菌分离株的多样性,并对这些代表株的抑菌活性进行初步探讨,以期筛选具有高效抑菌活性的鱼源益生菌。【方法】对分离的47株乳酸菌代表株进行p H、温度生长范围、耐盐性等生理生化特征检测,结合16S r RNA基因序列对已分离到的乳酸菌进行基因分型和菌种鉴定,采用牛津杯双层平板法检测乳酸菌代表株的抑菌活性。【结果】鉴定结果显示:23株为Lactobacillus fuchuensis(48.94%),12株为Lactobacillus curvatus(25.53%),3株为Leuconostoc fallax(6.38%),2株为Lactobacillus sakei(4.26%),2株为Weissella ceti(4.26%);2株为Lactococcus cremoris(4.26%),1株为Leuconostoc lactis(2.13%),1株为Weissella minor(2.13%),1株为Enterococcus devriesei(2.13%)。qz1217、qz1196、qz1220所在的A、B、C三组乳酸菌在5-50°C的温度范围内生长良好,qz1196、qz1220所在的B、C组在pH 3.0-10.0的范围内生长良好,几乎所有乳酸菌都具有耐6.5%盐浓度特性。13株乳酸菌菌株对6种病原菌都具有抑制作用。通过排除酸、过氧化氢实验,发现上清液仍然具有抑菌活性。对qz1251发酵液进行蛋白酶处理,抑菌活性消失,确定其抑菌物质属于蛋白类物质,是一种细菌素。【结论】青海湖裸鲤肠道附着乳酸菌的多样性为益生性乳酸菌的筛选提供优质资源及数据参考。  相似文献

The complete mitochondrial DNA (mtDNA) cytochrome b gene (1140 bp) was sequenced in Herzenstein macrocephalus and Gymnocypris namensis and in 13 other species and sub-species ( n = 22), representing four closely related genera in the subfamily Schizothoracinae. Conflicting taxonomies of H. macrocephalus and G. namensis have been proposed because of the character instability among individuals. Parsimony, maximum likelihood and Bayesian methods produced phylogenetic trees with the same topology and resolved several distinctive clades. Previous taxonomic treatments, which variously placed these two species of separate genera or as sub-species, are inconsistent with the mtDNA phylogeny. Both H. macrocephalus and G. namensis appear in a well-supported clade, which also includes nine species of Schizopygopsis , and hence should be transferred to the genus Schizopygopsis . Morphological changes are further illustrated, and their adaptive evolution in response to the local habitat shifts during the speciation process appears to be responsible for conflicting views on the systematics of these two species and hence the contrasting taxonomic treatments. These species are endemic to the Qinghai-Tibetan Plateau, a region with a history of geological activity and a rich diversity of habitats that may have result in the parallel and reversal evolution of some morphological characters used in their taxonomies. Our results further suggest that speciation and morphological evolution of fishes in this region may be more complex than those previously expected.  相似文献

五条河流青海湖裸鲤的同工酶变异   总被引:2,自引:0,他引:2  
为全面了解青海湖裸鲤遗传多样性现状,采用水平淀粉凝胶电泳技术分析了青海湖裸鲤[样品采自青海湖的五条支流,分别为泉吉河(QJ)、沙柳河(SL)、哈尔盖河(HEG)、布哈河(BH)以及甘子河(GZ)]的13种同工酶,共记录26个位点,统计了基因频率,计算了其遗传多样性。结果表明:26个基因位点中多态位点6个,分别为Pgm、Hk、Cat、Gdh、Es-t1、S-Mdh,多态位点比例(P0.99)为23.08%,表明青海湖裸鲤的五个群体皆具有较高的遗传多样性水平;分别计算了五个群体的平均杂合度(H)为0.1161(HEG)、0.1178(GZ)、0.1304(SL)、0.1305(BH)、0.1287(QJ),多态位点比例(Ae)分别为1.3112(BH)、1.3069(SL)、1.2542(GZ)、1.2492(HEG)、1.2453(QJ);除泉吉河群体外,其它四个群体的遗传偏离指数均为负数,表明多数群体处于一定程度的杂合子缺失状态;计算了五个群体间的遗传距离以及遗传相似度,结果显示群体间遗传距离与遗传相似度与各个河流的地理位置具有很强的相关性;根据五个群体的遗传距离进行了聚类分析,通过构建系统树图将其基本分为两大种群,甘子河与哈尔盖河两群体为一类群,另外三个群体分为一个类群;计算群体间的基因流(Nm)为7.2013,说明河道之间具有较为广泛的基因交流,并计算了两两群体间的基因流,其中布哈河群体与其它群体间的基因交流都较为广泛。  相似文献

对藏北高原色林错湖泊中色林错裸鲤(Gymnocypris selincuoensis)的生长方程、生长拐点以及生长指标等生长特征进行了分析。结果表明von Bertalanffy生长方程、Gompertz方程和三次多项式方程都可以反映色林错裸鲤的生长过程,但Gompertz方程能够很好地描述12龄之前的生长特征,而von Bertalanffy生长方程更适合描述18龄以后的生长特征。这种情况反映了色林错裸鲤具有洄游性鱼类更换生活环境的生活史特征。雄性体长的von Bertalanffy生长方程为:Lt=484.1906[1-e^-0.06839(t-0.06028)],雄性为Lt=485.3285[1-e^-0.0710(t-0.5679)]。体长和体重关系为W=0.00023L^-5.5303(♂)和W=0.00046L^2.4072(♀)。生长拐点为12.9龄(♀)和14.2龄(♂)。与其它裂腹鱼类相比,色林错裸鲤的生长过程尤为缓慢,这与其生存的环境条件更为恶劣直接相关。  相似文献

在描述鳞片、背鳍条和矢耳石三种材料轮纹特征的基础上,比较了这些年龄鉴定材料在判读错鄂裸鲤年龄和反映生长特征上的异同。在个体早期生长阶段,耳石轮纹阻断、8龄以上个体鳞片上年轮环纹的缺失和背鳍条出现轮纹的重叠是影响错鄂裸鲤年龄准确判读的主要因素。采用耳石和鳞片的体长退算数据,Von Bertalanffy方程较好地描述了错鄂裸鲤的生长。由于背鳍条的生长在个体的生长过程中始终呈现为负的异速生长,因此耳石、鳞片在解释个体生长时优于鳍条。  相似文献

藏北3种裸鲤同工酶的电泳分析及物种分化的探讨   总被引:4,自引:0,他引:4  
对藏北高原3种裸鲤的乳酸脱氢酶(LDH)、苹果酸脱氢酶(MDH)和脂酶(EST)进行电泳分析的结果表明,3种裸鲤酶谱均表现出种间的差别,而且在同一种群个体之间也存在着明显的分化,但无性别差异。3种裸鲤被检测的3种同工酶均有沉默基因表达的现象,重复基因LDH-A^2、LDH-B^2、s-MDH-A^2和m-MDH-B^2也在部分个体中表达。遗传距离分析表明,色林错裸鲤(G.selincuoensis)与错鄂裸鲤(G.cuoensis)之间较之于与纳木错裸鲤(G.namensis)有更近的亲缘关系。与其他四倍体鱼类相比,裸鲤鱼类同工酶在重复基因和沉默基因上都有较高的表达频率,这种情况说明裸鲤鱼类目前可能还外于多倍化后进化的早期过程并早于胭脂鱼类所处的相应时期,这与裂腹鱼类起源较晚以及青藏高原业已存在的恶劣环境条件直接相关。  相似文献

青海湖裸鲤是青海湖唯一的经济鱼类,由于湖区生态环境的恶化,使得该鱼种群数量越来越稀少.2004年中国环境发展国际合作委员会在<中国物种红色名录>中,将其列为濒危物种.研究利用生物素-磁珠富集法获得10个具有多态的微卫星分子标记,检测6个野生裸鲤群体:布哈河群体、沙柳河群体、哈尔盖河群体、泉吉河群体、黑马河群体、淡水系群体的遗传多样性.结果表明:每个座位上的等位基因数为2~8个, 大小在161~486bp之间.有效等位基因数在1.00~5.45个, 平均多态信息含量分别为0.3642、0.4044、0.3830、0.4047、0.3869和0.3940, 无偏观测杂合度的平均值在0.4145~0.5105之间, 说明这几个群体属于中度多态, 遗传多样性水平较高.根据群体间的遗传距离和遗传相似系数发现,布哈河群体与淡水系群体之间的遗传距离最大(DA=0.1607),哈尔盖河群体与泉吉河群体之间的遗传距离最小(DA=0.0125).由UPGMA聚类图可知:哈尔盖河和泉吉河两个群体聚为一个分支,布哈河、沙流河、黑马河和淡水系4个群体聚为另一个分支.  相似文献

Gymnocypris przewalskii (Kessler 1876) is an endangered and state-protected rare fish species in Qinghai Lake, China. To further understand the life history and distribution of this fish, five surveys were carried out in Qinghai Lake between 2002–2006. Results of these surveys indicate that fishes were predominantly distributed about 2 m under the surface. In July, significant differences in fish density were found between surface and bottom layers (P = 0.001), and/or between middle and bottom layers (P = 0.025). Fish density was the greatest in the surface layer. In August and October, no significant differences were found between the different layers, but the bottom layer had a greater fish density. Furthermore, there were very large differences among different zones in fish distribution density. Differences in horizontal distribution were not significantly correlated to factors such as water depth and inshore distance, possibly because of very low and uniform fish density. Feeding, changes in water temperature, over-wintering and spawning appeared to influence fish distribution. Hydroacoustic estimates of G. przewalskii biomass in Qinghai Lake increased significantly between 2002 and 2006. We attribute this increase to the management measures put in place to protect this species.  相似文献

该研究比较观察青海湖裸鲤(Gymnocypris przewalskii)两亚种(青海湖裸鲤指名亚种(Gymnocypris przewalskii przewalskii)和青海湖裸鲤甘子河亚种(Gymnocypris przewalskii ganzihonensis))鳃的形态及显微结构。结果显示,指名亚种的第一鳃弓鳃耙数多、密集且较长,而甘子河亚种则耙数少、稀疏且短;两者的鳃丝间距及鳃小片间距无明显差异,指名亚种的鳃丝较长;指名亚种扁平上皮细胞形态以不规则椭圆形为主,鳃丝表面有黏液细胞,其上分布的氯细胞数量多于甘子河亚种,而甘子河亚种扁平上皮细胞的形态以六边形为主,鳃丝表皮未观察到黏液细胞。由此推测,青海湖裸鲤两亚种鳃形态的差异与其食性和栖息环境相适应。  相似文献
